Investigation of Anti-viral effect of SCOTIN and its derivatives
on Human Immunodeficiency Virus-1(HIV-1) production

There are a number of Drug Delivery System(DDS) being used to increase significantly the effect of drugs. Cell
Penetrating-Peptide(CPP) is one of DDS that can be applied for such purpose. Recently, a new Cell Penetrating-
Peptide designated as Advanced Cell penetrating Peptide(ACP) has been discovered in this laboratory and it has a
better cellular delivery activity than any other CPPs including a well-known CPP such as Tat. Previously, we and
others has demonstrated that SCOTIN expressed in the SHISA5 gene has a significant anti-viral effect against HCV
and HIV. In this study, we have examined whether anti-viral activity of SCOTIN against HIV-1 could be enhanced
and delivered by fusing the ACP. We have constructed various expression vectors of SCOTIN and ACP fusion SCOTIN.
Protein expression level and its location of each recombinant vectors were compared with SCOTIN. Then, we found
that APC fusion SCOTIN significantly reduced the viral level in the cell lysate and supernatants than over expression
of SCOTIN alone using co-transfection with a pNL4.3-GFP into 293T cell. These results suggested that ACP could
enhance the effect of anti-HIV-1 by SCOTIN. Thus, this approach might be useful for the development of a new
type of drug molecule against the HIV-1.
