Pig tissue factor pathway inhibitor α has better ability of
inhibiting blood coagulation caused by pig tissue factor than
human counterpart

Xenotransplantation is a good solution for the patients who meet with the shortage of volunteer’s organs. Xenograft
rejection caused by pig tissue factor (TF) is one of most severe problem for xenotransplantation. This rejection
induces severe inflammation with complement activation and infiltration of neutrophils. In this study, we invented
fusion protein with the constant region of immunoglobulin and tissue factor of human or pig (TFPI-Ig) to isolate
TFPI. PK15 cell lysate were used for analyzing TF activity downregulating ability of TFPI. Our results revealed that
pig TFPI has better therapeutic effects than human one. However, both TFPIs have cleavable site by some enzymes
in human blood. This study suggests a good possibility to block IBMIR caused in pig to human xenotransplantation
with pig TFPI-Ig.
