Inhibition of histone deacetylase 1/2 enhanced NK cell
mediated anticacenr immunity through induction of NKG2D
ligands

Histone acetylation regulates various ligand and genes expressions with epigenetic mechanisms. NKG2D Receptors
of immune cells are activated by natural killer group 2 (NKG2D) ligands. NKG2D ligand expression is upregulate as
histone deacetylases (HDAC) inhibitors. We predicted that HDAC genes to affect the expression of NKG2D ligands
and observed NKG2D ligands and major histocompatibility complexes (MHC) expressions in cancer cells treated
with HDAC1, HDAC2 inhibitors and specific small interfering RNA (SIRNA). Furthermore, treatment with selective
HDAC1/2 inhibitors was found to induce NKG2D ligand expression in transcription and proteomic. Cancer cells
susceptibility to NK cells was also increased. The increase in NKG2D ligand expression was also seen when HDAC1
or HDAC2 were silenced. These studies show that inhibiting HDAC1 and HDAC2 can induce expression of NKG2D
ligand and NK cell-mediated anticancer immunity improvement.
