The Circular RNA Expression Pattern after Intracerebral
Hemorrhage

Background: We investigated circular RNA (circRNA) expression pattern from a collagenase induced rat intracerebral
hemorrhage model and developed therapeutic strategy based on its expression.
Methods: Hemorrhagic stroke was induced by stereotactic collagenase injection and brain was harvested after 1, 3
and 7 days to study circRNA expression level by microarray platform and quantitative real time polymerase chain
reaction (qRT-PCR). The predicted target microRNA (miRNA) and parent messenger RNA (mRNA) levels of
significantly altered circRNAs were validated. Therapeutic strategy was determined based on the information about
circRNA, its potential target microRNA.
Results: The most elevated ones were rno_circRNA_002714 and rno_circRNA_002715, which are located closely each
other in chromosome 10, within exon sequence coding glial fibrillary acidic protein. The most significantly
downregulated circRNA was rno_circRNA_016465, which has complementary sequence with rno-miR-466b.
Conclusion: This study illustrated dynamic circRNA expression pattern in a hemorrhagic stroke model, which
correlated with miRNA and mRNA expression, suggesting its regulatory role of RNA dynamics in diseased condition
and supporting competing endogenous RNA hypothesis.
