Development and evaluation of triple layer skin phantom
model for continuous glucose monitoring system

Continuous glucose measurement system (CGMS) is actively researched for preventing hypoglycemic shock of
patients with type 1 diabetes by glucose monitoring in interstitial fluid. The aim of this study was to develop a triple
layer artificial skin phantom for evaluation of sensor insertion ability and physical characteristics of CGMS sensor.
Epidermis layer was fabricated by special silicone having a shore A value of 0.1 to 5, and dermis layer was fabricated
by 7%~40% gelatin. Subcutaneous fat layer was prepared by fixing with 0.5% agarose gel. Triple layer phantom,
which is composed of 15% gelatin-based shore A 0.3 silicon showed the most similar elasticity compared to under-
arm region as elasticity value of about 1.23±0.01 MPa, and 40% gelatin-based shore A 5 silicon showed the most
similar elasticity compared to under-arm region as elasticity value of about 1.82±0.01 MPa. Triple layer phantom,
which is composed of 40% gelatin-based shore A 0.3 silicon showed the most similar elasticity compared to human
mean elasticity as elasticity value of about 1.45±0.01 MPa. The triple layered artificial skin phantoms are expected
to utilize as an assessment tool in development of CGMS.
