The phytoestrogen component of flaxseed ameliorates

insulin resistance in B16BL6-induced muscle atrophy

Glucose intolerance is associated with cancer and insulin resistance, a major factor of impaired glucose tolerance,

can be an underlying cause of skeletal muscle loss. Previously, we reported the effect of SDG on the obesity, but
their role in cancer-related insulin resistance is largely unknown. Here, we investigated the regulatory mechanism

of SDG responsible for glucose disposal in cancer-bearing skeletal muscle. SDG showed significant reduction of
cancer weight in B16BL6 melanoma bearing mice. Moreover, SDG restored muscle weight of TA, accompanied by

reduction in the expression of muscle atrophy factor, muscle ring finger protein and muscle atrophy factor. Insulin
resistance in TA was improved by SDG administration, with both increased p-IRS ser107 and glucose transporter

type 4 (Glut-4). These results suggest that the in vivo insulin-sensitizing effects of SDG in cancer are in part
attributable to its direct action on skeletal muscle via downregulation of muscle atrophy and offer a new therapeutic

strategy for cancer-related insulin resistance.
