PLD1 has been implicated in a variety of inflammatory diseases, including arthritis, asthma, and cancer. However,
little is known about the role of PLD1 in vascular inflammation. RNAseq analysis showed a large decrease of PLD1

expression after the laminar flow of endothelial cells (ECs) without changes in other PLD isoforms. CRISPR/Cas9-
mediated PLD-1 gene knockout in ECs led to the blockade of disrupted flow-induced increased activity of NFkB. In

addition, it was confirmed that siPLD knockdown in ECs increased the activity of disturbed flow-induced NFkB, and
that the activity of Yap was also increased. These results suggest that PLD1 may mediate vascular inflammation and

        atherosclerosis by regulating EC mechanization.
